Depletion antibodies and genetically engineered mouse models are commonly used to investigate lymphocyte subsets in immunity, albeit with limitations. Kastner et al. develop adeno-associated viral vectors delivering depletion antibodies that eliminate lymphocyte subsets long-term. Leveraging this strategy, they interrogate how B cells and CD8+ T cells control chronic viral infection.
