Mashup Score: 42Agnostic drug development revisited - 3 day(s) ago
The advent and rapid implementation of tissue-based or circulating tumor DNA next generation sequencing (NGS) in the clinic has enabled the identification of novel actionable genomic alterations present across tumor types[1]. Moreover, the number of agents approved that target specific actionable alterations has also increased enabling a shift in cancer treatment towards more personalized approaches paving the way for the implementation of precision oncology[2]. To date, a number of agents that target specific actionable alterations have shown superiority over standard-of-care cytotoxic agents and are part of the treatment armamentarium in a variety of cancers[3–6].

Mashup Score: 86How Humble Leadership Really Works - 2 month(s) ago
Top-down leadership is outdated and counterproductive. By focusing too much on control and end goals, and not enough on their people, leaders are making it more difficult to achieve their own desired outcomes. The key, then, is to help people feel purposeful, motivated, and energized so they can bring their best selves to work. One of the best ways is to adopt the humble mind-set of a servant leader. Servant leaders view their key role as serving employees as they explore and grow, providing tangible and emotional support as they do so. They actively seek the ideas and unique contributions of the employees that they serve. This is how servant leaders create a culture of learning, and an atmosphere that encourages followers to become the very best they can.

PURPOSE Recent evidence has shown that higher tumor mutational burden strongly correlates with an increased risk of immune-related adverse events (irAEs). By using an integrated multiomics approach, we further studied the association between relevant tumor immune microenvironment (TIME) features and irAEs. METHODS Leveraging the US Food and Drug Administration Adverse Event Reporting System, we extracted cases of suspected irAEs to calculate the reporting odds ratios (RORs) of irAEs for cancers treated with immune checkpoint inhibitors (ICIs). TIME features for 32 cancer types were calculated on the basis of the cancer genomic atlas cohorts and indirectly correlated with each cancer’s ROR for irAEs. A separate ICI-treated cohort of non–small-cell lung cancer (NSCLC) was used to evaluate the correlation between tissue-based immune markers (CD8+, PD-1/L1+, FOXP3+, tumor-infiltrating lymphocytes [TILs]) and irAE occurrence. RESULTS The analysis of 32 cancers and 33 TIME features demonstra
