Background Cervical cancer, encompassing squamous cell carcinoma and endocervical adenocarcinoma (CESC), presents a considerable risk to the well-being of women. Recent studies have reported that squalene epoxidase (SQLE) is overexpressed in several cancers, which contributes to cancer development. Methods RNA sequencing data for SQLE were obtained from The Cancer Genome Atlas. In vitro experiments, including colorimetry, colony formation, Transwell, RT-qPCR, and Western blotting were performed. Furthermore, a transplanted CESC nude mouse model was constructed to validate the tumorigenic activity of SQLE in vivo. Associations among the SQLE expression profiles, differentially expressed genes (DEGs), immune infiltration, and chemosensitivity were examined. The prognostic value of genetic changes and DNA methylation in SQLE were also assessed. Results SQLE mRNA expression was significantly increased in CESC. ROC analysis revealed the strong diagnostic ability of SQLE toward CESC. Patient

Background Several studies reported the sterol ester (SE), a subclass of subtype of cholesterol ester (CE), is associated with the incidence of colorectal cancer (CRC). Nevertheless, the causal relationship of SE on CRC remains unknown. Methods A two-sample Mendelian randomization study was performed with the summary statistics of SE (27:1/14:0) which is from the largest available genome-wide association study meta-analysis(n = 377277) conducted by FinnGen consortium. The summary data were obtained from UK Biobank repository (377673 cases and 372016 controls). And we used a relative relaxed filter (p < 5 × 10− 6 and LD r2 < 0.01) of instrumental variables to explore the causal effect and complete the sensitive analysis with the threshold p < 5 × 10− 8 and LD r2 < 0.01, MR Egger intercept, MR-PRESSO, and leave-one-out method, which all support the causal assessment. Inverse variance weighted, MR-Egger, weighted median, simple mode, and weighted model, were used to examine the causal ass

Mashup Score: 0ILF2 protein is a promising serum biomarker for early detection of gastric cancer - BMC Cancer - 1 month(s) ago
Background Our previous small-sample study indicated that serum levels of interleukin enhancer binding factor 2 (ILF2) may have the potential for gastric cancer (GC) detection. The present study was conducted to further validate the diagnostic value of serum ILF2 protein for GC. Methods Serum specimens and clinical data were collected from patients with GC (n = 99) or benign gastric disease (BGD) (n = 49) and healthy controls (HC) (n = 51). Serum ILF2 levels were measured using enzyme-linked immunosorbent assay. The diagnostic performance of ILF2 was evaluated using the area under the receiver operating characteristic curve (AUC). The independence and synergy of ILF2 in GC diagnosis were analyzed by modeling with conventional blood indicators. Results The median serum ILF2 level was higher in the GC group (227.8ng/mL) than in the BGD group (72.0ng/mL) and the HC group (56.8ng/mL) (p < 0.001), and no significant difference across GC subgroups. The AUCs of ILF2 were 0.915 (95%CI 0.873–0.

Background Accurate diagnosis and therapeutic response to secondary central nervous system lymphoma (SCNSL) are challenges that need to be addressed. We assessed the value of cerebrospinal fluid (CSF) cytokine levels for diagnosis and post–therapeutic prognosis in patients with SCNSL. Methods This retrospective study included 234 patients with non-Hodgkin lymphoma (NHL), including the SCNSL group (n = 57) and the non-SCNSL group (n = 177). The Mann–Whitney U test was used to compare the cytokine profiles between SCNSL and non-SCNSL group. Receiver operating characteristic curve was used to determine the diagnostic ability of CSF cytokine levels for SCNSL. The predictive value of CSF cytokine concentrations for progression free survival of patients with SCNSL was evaluated using a log-rank test. Results CSF IL-6 and IL-10 levels in SCNSL group were significantly elevated compared with those in the other two groups. ROC curve showed that the cutoff values of IL-6 and IL-10 in CSF were 10

Objective To compare the performance of [18F]FDG and [18F]FAPI-04 in PET/CT evaluation for liver cancer lesions, with a further exploration of the associations between PET semiquantitative data and immunohistochemical markers to liver cancer. Methods Patients with suspected malignant liver lesions (MLL) underwent [18F]FDG and [18F]FAPI-04 PET/CT scanning. Liver lesions were visually classified as positive or negative based on their uptake level exceeding that of adjacent normal liver tissue. SUVmax and tumor-to-background ratio (TBR) were recorded for semi-quantitative analysis. Sensitivity, specificity and accuracy of each tracer were determined using pathological findings as the gold standard. Furthermore, immunohistochemical analysis provided the molecular characteristics of all MLLs. Comprehensive analysis explored correlations between these molecular markers and PET semiquantitative parameters (SUVmax andTBR) to identify potential associations. Results The study enrolled 44 patien

Mashup Score: 0RAI therapy in low-risk papillary thyroid cancer: recurrence reduction and long-term outcomes in the Turkish population - BMC Cancer - 3 month(s) ago
Purpose Papillary thyroid cancer (PTC) is the most common thyroid malignancy, characterized by its slow progression and favorable prognosis. This study re-evaluates the efficacy of radioactive iodine (RAI) therapy versus no RAI in low-risk PTC patients following total thyroidectomy. Methods A retrospective analysis was conducted on 588 patients treated between 2010 and 2016 at a major tertiary center in Turkey. Patients were divided into two cohorts: those receiving total thyroidectomy (TT) with high-dose RAI (100 mCi) and those receiving TT alone. A matched cohort of 138 patients per group was analyzed to minimize bias. Results Follow-up data indicated that at 24 months, the RAI group demonstrated a higher percentage of excellent treatment responses (86%) compared to the non-RAI group (74%). Long-term follow-up showed that 99.3% of the RAI group achieved no evidence of disease (NED), versus 90.6% in the non-RAI group. Recurrence rates were significantly lower in the RAI group (1%) com
