Uveitis is a heterogenous group of inflammatory eye disease for which current cytokine-targeted immune therapies are effective for only a subset of patients. We hypothesized that despite pathophysiologic nuances that differentiate individual disease states, all forms of eye inflammation might share common mechanisms for immune cell recruitment. Identifying these mechanisms is critical for developing novel, broadly acting therapeutic strategies.

Immune checkpoint inhibitors (ICIs) and MAP-kinase pathway inhibitors (MAPKis) are the most common drugs responsible for drug-induced uveitis, according to a study of the World Health Organization global pharmacovigilance database, Vigibase.1 Immune checkpoint inhibitor drug-induced uveitis seem to be more frequent in patients treated for malignant melanoma, suggesting that melanin-associated proteins released during melanoma cell lysis may promote dysregulation of inflammation.2 MAPK inhibitors include 2 classes of drugs, BRAF inhibitors (BRAFis) and MEK inhibitors (MEKis), which are mainly used to treat unresectable or metastatic melanoma, either alone or in combination, and are involved in several types of ocular side effects including ocular inflammatory disorders.
