In this large study, published in JCO, the use of high dose methotrexate was not associated with a clinically meaningful reduction in risk of CNS progression.

This recent study in JHO highlighted outcomes of patients with secondary CNS lymphoma who received CAR T-cells. The median PFS was just 3.3 months (95% CI = 2.6–6.0 months) with 6- and 12-month PFS rates of 35% and 16%, respectively.

While this one isn't CNS lymphoma themed, it is a must-read. Gloria Iacoboni and colleagues demonstrate that patients who have received bendamustine prior to apheresis for CAR T-cells have worse outcomes than those not exposed to benadmustine. Seems it is worth avoiding bendamustine prior to CAR T-cell use.
